Dr. Pluznick discovered that olfactory receptors in mice are also expressed in their kidneys and blood vessels. Her research is focused on the role of chemosensory GPCRs in regulating renal and cardiovascular function, and identifying renal/cardiovascular olfactory receptor ligands, and relating them to whole-animal physiology. This work contributes to a better understanding of how the kidney helps maintain homeostasis in humans. Jennifer is currently an assistant professor of physiology at the Johns Hopkins School of Medicine. She received her undergraduate degree in biology from Truman State University and earned her Ph.D. in renal physiology from the University of Nebraska Medical Center. She then spent five years training as a postdoctoral fellow in the laboratory of Michael Caplan at Yale University, where she studied both renal physiology and sensory biology systems and focused on olfaction. | |||

Duke Professor of Medicine and Professor of Biochemistry, Chemistry, and Pathology at the Duke University Medical Center. He began his career in the late 1960s and has been an Investigator of the Howard Hughes Medical Institute since 1976. His legacy lies in the numerous discoveries he and his team made in the GPCR field and in all those who trained in his laboratory and went on to pursue stellar scientific careers. Dr. Kathryn E Livingston is currently a Product Manager at Gator Bio, a biotechnology company providing solutions to researchers studying protein-protein interactions. Kathryn obtained her BS in Chemistry from Carnegie Mellon University and went on to receive a Ph.D. in Pharmacology at the University of Michigan. Working under the supervision of Dr. John Traynor, Kathryn worked to develop and understand first-in-class allosteric modulators of opioid receptors. Research into their mechanism of action in purified systems formed the base of her thesis work. Following this, Kathryn did a post-doctoral fellowship at UCSF in the laboratory of Dr. Mark von Zastrow. There she researched the beta-2 adrenergic receptor and developed assays to investigate real-time activity in intact cellular systems. Dr. Maria Waldhoer is originally from Austria. She earned her M.Sc. in Zoology and Neurobiology before completing a Ph.D. in Biology and Pharmacology at the University of Vienna. GPCRs led Maria to Thue W. Schwartz’s lab in Copenhagen where she completed her postdoctoral training. After working in the US and at the University in Graz in Austria, Maria worked several years at Novo Nordisk before joining InterAx Biotech in Switzerland as their Chief Scientific Officer. Even though Maria stumbled upon the GPCR field, her 20 years in both academia and in the industry working on GPCRs make her a strong and dedicated scientific leader. | |||

Oprea , Thomas Sakmar, Aurelien Rizk & Yaroslav Nikolaev. The explosion of biomedical data such as in genomics, structural biology, and pharmacology can provide new opportunities to improve our understanding of human physiology and disease. In recent years, machine learning (ML) and artificial intelligence (AI) methods have received a significant boost of attention. ML/AI can be powerful for identifying abstract patterns within large data where traditional methods would be oblivious to. This comes without the need for manual feature engineering as systems can learn through implicit rules from the data provided. G protein-coupled receptors (GPCRs) mediate a vast variety of critical biological processes and provide an ideal case study for quantitative, and multi‐scale integration of these amounts of data to gain novel insights into receptor biology. How can we best leverage these exciting new techniques in areas such as protein structure prediction, bioactive ligand discovery, in vivo translationability or in our understanding of signalling determinants? Dr. Chris Tate obtained his Ph.D. from the University of Bristol in 1989 and then moved to the University of Cambridge (Dept of Biochemistry) to work on bacterial sugar transporters. In 2005 he started working on the development of conformational thermostabilization of GPCRs, which resulted in the structure of the β1-adrenoceptor. In 2016 mini-G proteins were developed as a tool for the structure determination of GPCRs in the fully active state. At the end of his studies, he was awarded the Pfizer Prize for undergraduate research. He then completed his Ph.D. at Leeds as well as the University’s Ackroyd, Brotherton, and Brown Scholar. Following his Ph.D., Dr. Maudsley was awarded a Howard Hughes Medical Institute Fellowship to train with Professor Robert Lefkowitz at Duke University. Following this tremendous experience, he was recruited to be the Principal Investigator of the Receptor Biology Section at the Medical Research Council (MRC) -Human Reproductive Sciences Unit within the University of Edinburgh. At the MRC he developed novel prostate cancer therapeutics based upon his research into GPCR pluridimensional signaling. To broaden his biomedical skill-set Stuart next accepted the position of Head of the Receptor Pharmacology Unit at the National Institutes of Health – National Institute on Aging at the Johns Hopkins University Medical Center. At the NIH he was the recipient of the coveted NIH ‘Bench-to-Bedside’ Translational Research Grant Award, one of the few awards available within the intramural NIH program. Upon starting a new family, and returning to Europe, Dr. Maudsley continued his scientific journey with the award of the highly-valued Odysseus Program Type I Program Grant to work as both the Adjunct Director of the VIB Center for Molecular Neurology and also Vice-Chair of the Department of Biomedical Sciences at the University of Antwerp. Stuart’s current research, in the Receptor Biology Lab, focuses on the development of novel GPCR-based therapeutics that interdict diseases based on their gerontological underpinnings. In this episode of the Dr.GPCR podcast, my guest is Dr. Arun Shukla from the Indian Institute of Technology in Kanpur, India. Arun is currently an Associate Professor & Joy Gill Chair Professor, Intermediate Fellow, Wellcome Trust DBT India Alliance Swarnajayanti Fellow & EMBO Young Investigator at the Department of Biological Sciences and Bioengineering. He earned his master's degree in biotechnology from Jawaharlal Nehru University in India and it was during a biochemistry class where he learned about cell signaling that he became curious and wanted to learn more about it. Arun first started working on GPCRs and their structural characterization at the Max Planck Institute of Biophysics where he completed his doctoral studies in the lab of Dr. Hartmut Michel. Fascinated by GPCRs he wrote to Dr. Bob Lefkowitz and asked him if he could join his lab at Duke University. Dr. Shukla spent several years in the Lefkowitz lab and collaborated extensively with Dr. Brian Kolbika of Stanford University. I then obtained an M.Sc. at the University in Munich working e.g. in the lab of Patrick Cramer. I next moved to Rockefeller University in NYC to work in the lab of the late Dr. Guenter Blobel, before starting my Ph.D. in 2009 at The Scripps Research Institute in La Jolla. There I obtained my Ph.D. in the lab of Dr. Ray Stevens in 2013 solving several GPCR crystal structures, including that of the first serotonin receptor. I then moved to UNC at Chapel Hill to do postdoctoral work in the lab of Dr. Bryan Roth where I established GPCR structural biology and learned the ins and outs of molecular pharmacology and in vitro drug discovery. In 2018 I started my own lab at the Icahn School of Medicine at Mount Sinai in NYC, where I have been working on structure-function and drug discovery of GPCRs and transporters. Brian has contributed to the discovery or development of 5 marketed drugs through his work spanning molecular, biochemical, cellular, and in vivo assessment of drug candidates in many different physiological systems. Dr. Arey’s laboratory discovered the first described synthetic agonists and antagonists of the FSHR and has been an early champion of signaling bias as a physiological mechanism of gonadotropin action. Michel Bouvier is a professor of Biochemistry and Molecular Medicine and the CEO of the Institute for Research in Immunology and Cancer (IRIC) at the Université de Montréal. Following his Ph.D. in Neurological Sciences at the same university in 1985, he completed a post-doctoral fellow at Duke University in the laboratory of R.R. Lefkowitz. In 1989, he returned to Montréal as a professor of biochemistry and a scholar of the Medical Research Council of Canada at the Faculty of Medicine of the Université de Montréal. Since 2001, he holds the Canada Research Chair in Signal Transduction and Molecular Pharmacology. Dr. Bouvier is the author of 300 scientific papers and 15 patents and delivered close to 500 invited conferences. He is a world-renowned expert in the field of cell signaling and GPCRs and made seminal contributions to our understanding of this major class of drug targets. In addition to paradigm shifts including inverse agonism, biased signaling, and pharmacological chaperones, his work on bioluminescence resonance energy transfer (BRET) resulted in the development of screening assays that are now widely used for drug discovery. His work received more than 30,000 citations yielding an h-index of 95. He has supervised the research work of 75 graduate students and 40 post-doctoral fellows. Michel’s scientific contributions were recognized by the attribution of many awards and distinctions including his election as a fellow of the Royal Society of Canada (2014), the Julie Axelrod award from the American Society of Pharmacology and Exerimental Therapeutics (2017), the Wilder Penfield award from the Quebec Government (2017), the innovation award of ADRIQ (2019) and the 2021 Killam prize form the Canada Council for the Arts. ------------------------------------------- Imagine a world in which the vast majority of us are healthy. Dr. Pina Cardarelli, CSO for GPCR Therapeutics Inc., based in South Korea, has recently been named President of GPCR Therapeutics, USA, a newly incorporated Biotechnology company in the Bay Area. The company’s mission is to discover and develop highly effective cancer therapeutics by targeting heteromers of G protein-coupled receptors (GPCR). Burixafor, their most advanced clinical candidate, will be in Phase II clinical trial next year. Additionally, they have a library of target GPCR heteromers for Oncology. Dr. Cardarelli heads the team of talented researchers that will be expanding at the US site. Dr. Cardarelli is a drug development leader with extensive experience driving drug discovery teams in bringing biologics to clinical proof of concepts. She has expertise in cell biology, pharmacology, translational medicine, oncology, immuno-oncology, immunology, and clinical development. Previously, she held the position of Vice President of Cell Biology & Pharmacology, at Bristol-Myers Squibb. She was an integral contributor to two therapeutics that are FDA approved, Yervoy and Opdivo. She was a participant in numerous due diligence (anti-CXCL8 mAb) and has managed external collaborations and alliances. Prior to this, she held the position of Vice President, at Medarex, Inc. While at BMS and Medarex, she led programs from target ID to clinical development that included, CXCL10 (Eldelumab), CXCR4 (Ulocuplumab), CD30, CD19, Fucosyl GM-1, & mesothelin-ADC, Glypican-3-ADC, CD70-ADC. She oversaw early discovery programs IL-23 p19 and IL23 p19/IL-17 bispecifics. At Medarex, she initiated and identified the lead mAb for the type I interferon-alpha receptor project, licensed to AstraZeneca (Saphnelo™ Anifrolumab) that has just received FDA approval for systemic lupus erythematosus. She has extensive experience working with Biologics, and Antibody Drug Conjugates as well as experience in IND fillings, IB updates, and responding to FDA inquiries. She is an inventor on 39 issued U.S. patents including anti-PD-1 patents, 22 EP patents, and greater than 100 global patents centered around therapeutic development. She has also authored forty-six peer-reviewed publications. Brian Arey is Senior Director of Mechanistic Pharmacology within Leads Discovery and Optimization at Bristol-Myers, Squibb Co. in Lawrenceville, NJ. He obtained both his MS and Ph.D. in Neuroendocrine Physiology at Florida State University before completing his postdoctoral training at Northwestern University. He then moved to work in the pharmaceutical industry where he has held positions of increasing responsibility. He currently leads a team that provides a mechanistic understanding of small molecule drug candidates across the entire portfolio of BMS. Brian has contributed to the discovery or development of 5 marketed drugs through his work spanning molecular, biochemical, cellular, and in vivo assessment of drug candidates in many different physiological systems. Dr. Arey’s laboratory discovered the first described synthetic agonists and antagonists of the FSHR and has been an early champion of signaling bias as a physiological mechanism of gonadotropin action. He continues to pioneer in drug discovery studying GPCRs and other target classes. His recently published book on signaling bias, Biased Signaling in Physiology, Pharmacology, and Therapeutics is available on Amazon. I sat down with Brian to chat about GPCRs, working in the industry, and being a leader. Adriano received his Bachelor of Science degree in Pharmacology in 1991 from the University of Toronto. He then moved to Thomas Jefferson University in Philadelphia, PA, for his postdoctoral training in Jeff Benovic’s laboratory studying the regulation of G protein-coupled receptor trafficking and signaling. In 2004 Adriano joined the faculty of the Department of Pharmacology at Loyola University Chicago. ''Dr. Ross Cheloha is an Investigator at the National Institutes of Health in the Laboratory of Bioorganic Chemistry in Bethesda, MD, where he started in October 2020. He completed his postdoctoral training at MIT and Harvard Med School in the lab of Hidde Ploegh where he developed new applications of single domain antibodies (nanobodies). He earned his PhD in Chemistry at the University of Wisconsin-Madison in the lab of Sam Gellman on the study of analogs of the GPCR peptide ligand parathyroid hormone. Work in his independent laboratory is focused on developing new pharmacological tools via chemistry and protein engineering to interrogate GPCR signaling. Sam completed his Ph.D. in biochemistry, studying allosteric modulation of dopamine receptors, from the University of Kent, United Kingdom. He then moved to the National Institute of Mental Health, researching pharmacological mechanisms of Class B GPCRs as part of his postdoctoral training. Today, Sam is a pharmacology data analyst and the founder of Pharmechanics LLC, a consultancy and data analysis company supporting pharmaceutical, life science, and academic scientists in the development of new therapeutics and the understanding of receptor systems. As an industry pharmacologist, he consults with numerous pharma and biotechs in understanding and applying in vitro pharmacology data to advance drug discovery. He specializes in kinetic analysis of drug action and is known for applying binding kinetics to the development of effective therapeutics, particularly GPCR antagonists. Before founding Pharmechanics in 2016, Dr. Hoare was a pharmacology leader in the pharmaceutical industry for 15 years at Neurocrine Biosciences. He guided the in vitro biology efforts of the company for numerous drug discovery campaigns. Sam is known for demystifying complicated and newly-emerging pharmacology concepts, enabling them to be applied by project teams in optimizing new molecules. In this science-focused episode of the Dr. GPCR podcast, we meet Dr. Ian Chronis, a freshly minted PhD from the University of Michigan. While his academic roots started in classical history and ancient Greek, Ian’s curiosity pulled him toward chemistry, then pharmacology, and eventually into the complex world of GPCR signaling. His story is a testament to the power of staying open to change and following emerging scientific questions wherever they lead. Ian’s doctoral work focused on two key GPCR systems: the well-characterized β2-adrenergic receptor and the lesser-known proton-sensing receptor GPR65. In the former, he explored how PKA phosphorylation modulates receptor interactions and discovered a regulatory role for 14-3-3 proteins. But it’s GPR65 — a receptor that constitutively internalizes and signals from acidic endosomes — that has captured Ian’s attention and become the centerpiece of his current research. Unlike classical GPCRs, GPR65 uses non-canonical internalization mechanisms, presenting new opportunities to uncover novel biology. The receptor’s ability to signal within the tumor microenvironment and immune cells makes it an emerging drug target, especially in cancer immunotherapy. However, there are still major gaps: no selective ligands, little clarity on its internalization machinery, and limited tools to modulate its function. Ian’s wish list? A high-affinity agonist and antagonist to begin untangling these questions. Until then, he's carefully optimizing pH-based assays and preparing for next steps in a postdoctoral lab focused on more physiological and therapeutic systems. Whether he’s decoding novel trafficking pathways or reflecting on career strategy, Ian brings a balanced perspective that’s both ambitious and grounded. His advice to peers? Attend talks outside your field, learn constantly, and surround yourself with people who make science sustainable. About Dr. Sudha ShenoyDr. Sudha Shenoy is currently an Associate Professor in Medicine & Cell Biology in the Division of Cardiovascular Medicine, Duke University Medical Center. She received her Ph.D. from Oklahoma State University and completed her postdoctoral training with Dr. Robert J. Lefkowitz (Nobel Laureate, 2012) at Duke University. Dr. Shenoy’s postdoctoral research discovered that ubiquitination of mammalian G protein-coupled receptors is a tag for lysosomal degradation, whereas ubiquitination of the adaptor protein, β-arrestin, is a tag for receptor internalization and formation of signaling endosomes. Her laboratory has continued to work on identifying the molecular mechanisms that ascribe ubiquitin code on GPCRs and β-arrestins. Fiona Marshall got fascinated with GPCRs after attending a lecture on how the beta-adrenergic receptor in the heart is activated by adrenaline, during her undergraduate studies at Bath University. She then pursued her Ph.D. in neuroscience from Cambridge University. An expert in GPCR biology, Fiona published the first description of the cloning and structural requirements of the GABAB receptor. One of her career path defining moments came from when she visited Dr. Chris Tate and Dr. Richard Henderson at the Laboratory of Molecular Biology in Cambridge, UK. As a co-founder of Heptares Therapeutics, now called Sosei Heptares, a GPCR focused drug discovery and development biotechnology company, Fiona and her team made considerable breakthroughs in the field of GPCR stabilization and structure-based drug design. Today, Dr. Marshall is the VP Head of Neuroscience Discovery and Head of Discovery UK, Global Head of Neuroscience discovery research leading teams in West Point, Boston, and London at MSD. Stuart graduated from the University of Leeds in the U.K. with a First Class Honors degree in Pharmacology. At the end of his studies, he was awarded the Pfizer Prize for undergraduate research. He then completed his Ph.D. at Leeds as well as the University’s Ackroyd, Brotherton, and Brown Scholar. Following his Ph.D., Dr. Maudsley was awarded a Howard Hughes Medical Institute Fellowship to train with Professor Robert Lefkowitz at Duke University. Following this tremendous experience, he was recruited to be the Principal Investigator of the Receptor Biology Section at the Medical Research Council (MRC) -Human Reproductive Sciences Unit within the University of Edinburgh. At the MRC he developed novel prostate cancer therapeutics based upon his research into GPCR pluridimensional signaling. To broaden his biomedical skill-set Stuart next accepted the position of Head of the Receptor Pharmacology Unit at the National Institutes of Health – National Institute on Aging at the Johns Hopkins University Medical Center. At the NIH he was the recipient of the coveted NIH ‘Bench-to-Bedside’ Translational Research Grant Award, one of the few awards available within the intramural NIH program. Upon starting a new family, and returning to Europe, Dr. Maudsley continued his scientific journey with the award of the highly-valued Odysseus Program Type I Program Grant to work as both the Adjunct Director of the VIB Center for Molecular Neurology and also Vice-Chair of the Department of Biomedical Sciences at the University of Antwerp. Stuart’s current research, in the Receptor Biology Lab, focuses on the development of novel GPCR-based therapeutics that interdict diseases based on their gerontological underpinnings. This research stream is now forming the basis of a new technology-based start-up company, HeptOME, to help screen and develop novel longevity/disease-regulating compounds with multidimensional disease efficacy profiles. ------------------------------------------- Imagine a world in which the vast majority of us are healthy.
